No, it is immediate. You may need to do a browser refresh, but there is no delay.

So if you're not seeing the locations, I wonder if your stake clerks understood that they need to check the box for "This location can be scheduled by Wards and Stakes"

I vote for a sticky of this thread with the initial post edited to include this info (or at least a rewording that reading the administrators guide link is imperative...)
